Immune Checkpoint Functional Assays

Equipped with professional technical scientists, powerful platforms, and abundant experience, Creative Biolabs provides comprehensive immune checkpoint functional assays to facilitate our clients' meaningful immunotherapy project development. We offer high-quality custom services by adjusting strategies to meet particular requirements.

Immune checkpoints maintain self-tolerance and help to avoid collateral damage during a physiological immune response. Studies have shown that tumors engineer microenvironments to evade immune surveillance and attack, particularly by modulating specific immune checkpoint pathways. Recently, immune checkpoint therapies, particularly in monoclonal antibodies, have been intensely researched and deployed as novel immunotherapy agents to treat cancers. Therefore, immune checkpoint functional assays are highly needed to understand the mechanism of immune checkpoint blockade therapy.

Immune Checkpoint Functional Assays at Creative Biolabs

  • T cell proliferation assay
  • Creative Biolabs has accumulated extensive experience in conducting T cell proliferation assays to validate the anti-tumor activity of immuno-oncology drugs. Our scientists have developed a systematic strategy for T cell proliferation assays for immuno-oncology drug discovery. Our strategy consists of bioluminescent-based methods, ELISA, carboxyfluorescein succinimidyl ester (CFSE) proliferation assay, methyl thiazol-diphenyl-tetrazolium bromide (MTT) assay and flow cytometric analysis, etc. We provide custom T cell proliferation assays to facilitate our clients' research and projects.

  • Cytokine production/release assay
  • Creative Biolabs offers high-quality cytokine release assays, which can be customized to meet the demands of our clients' programs, using primary human or non-human primate (NHP) cells, sourced fresh to ensure proximity to the state of circulating cells in vivo. The high sensitivity of our cytokine release assay allows rapid and reliable quantification of the production of multiple cytokines, such as granulocyte colony-stimulating factor (G-CSF), granulocyte/macrophage colony-stimulating factor (GM-CSF), Interferon-gamma (IFN-γ), Interleukin (IL)-1β, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-10, IL-12p70, IL-13, IL-17A, MCP-1, macrophage inflammatory protein 1 beta (MIP-1β) and tumor necrosis factor α (TNFα). Multiple therapeutic antibodies associated with cytokine release-mediated clinical toxicity can be used to correlate the relative severity of the cytokine response induced by the test products.

  • T cell cytotoxicity assay
  • Real-time, kinetic measurement of T cell cytotoxicity offers a wealth of information in a label-free manner and becomes the preferred method for measuring cytotoxicity. Creative Biolabs provides T cell cytotoxicity assay in a label-free, real-time way. Our services include long-term cytotoxicity assays for revealing T cell killing in cell cultures and compound-mediated cytotoxic effects. Our T cell cytotoxicity assays can expedite your candidate selection process.

  • Mixed lymphocyte reaction (MLR) assay
  • MLR is a powerful way to measure the effectiveness of your therapeutic candidate on T cell activation. Depending on your research question, Creative Biolabs offers one-way MLR (T cells from one donor are mixed with antigen-presenting cells from an alternative donor) and two-way MLR (T cells from both donors are combined, each effectively containing both responder and stimulator cells). Moreover, our seasoned scientists can repeatedly use the same donor pairs or test numerous donor pairs.

  • In vitro drug resistance studies
  • Creative Biolabs is well equipped and versed in in vitro drug resistance studies. Our scientists have accumulated rich experience in a range of in vitro techniques, including human tumor cloning assay (HTCA), thymidine incorporation assay (TIA), adenosine triphosphate bioluminescence (ATP-b) assay, MTT assay, differential staining cytotoxicity (DISC) assay, and extreme drug resistance (EDR) assay.

Creative Biolabs has organized a staff of outstanding scientists who have engaged in immune checkpoint functional studies for many years. Integrating state-of-the-art technologies as well as industry-leading expertise, we offer a comprehensive panel of immune checkpoint functional assays. Our scientific team has the necessary capabilities to provide overall immune checkpoint functional study solutions, focusing on innovative research.
